What is Semax?

Semax is a synthetic heptapeptide analogue derived from the ACTH (4–10) fragment, containing the sequence Met–Glu–His–Phe–Pro–Gly–Pro (MEHFPGP). Originally developed in Russia during the 1980s, it was engineered to enhance metabolic stability and neuroactive properties—effectively bypassing the hormonal activity of full-length ACTH while retaining central nervous action.

Chemical Structure of Semax

Semax’s primary structure consists of the seven amino acids Met–Glu–His–Phe–Pro–Gly–Pro, with chemical formula C₃₇H₅₁N₉O₁₀S and an average molecular weight of 813.9 Da, as confirmed by PubChem (CID 122178). It incorporates a C-terminal Pro‑Gly‑Pro tripeptide (PGP) to enhance resistance to enzymatic degradation, ensuring prolonged neuropeptide activity in experimental models.

What Are the Effects of Semax?

  • Neuroprotective & Gene Expression Modulation: In rodent stroke models, Semax reduces inflammatory gene expression (e.g., MMP-9, c-Fos, JNK), upregulates CREB and BDNF, and supports neuron survival in ischemic penumbra.
  • Cognitive Enhancement & BDNF Elevation: A single dose significantly increases BDNF protein and mRNA in the hippocampus (~1.4×), with corresponding gains in learning and memory performance in preclinical models.
  • Monoaminergic System Support: Studies report Semax enhances dopamine and serotonin transmission in the striatum and cortex, facilitating neurobehavioral resilience.
  • Immunomodulation & Angiogenesis: In ischemic rodent models, Semax shifts expression of immune-regulating chemokines and immunoglobulins, and supports angiogenic gene expression to promote vascular recovery.
  • Nootropic & Default Mode Network Effects: Functional MRI in humans demonstrates Semax alters default mode network activity, linked to enhanced cortical connectivity and attentional processing.
