SS-31 (Elamipretide)

SS-31 is a synthetic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2, containing the non-natural amino acid 2,6-dimethyltyrosine and a D-arginine residue at the N-terminus. Its alternating aromatic-cationic structure allows it to concentrate in the inner mitochondrial membrane, where it associates with cardiolipin, a phospholipid specific to that membrane. In published preclinical literature, SS-31 has been characterized in connection with cardiolipin binding, cristae architecture, and electron transport chain function. SS-31 has been evaluated in clinical trials for primary mitochondrial myopathy and Barth syndrome.
