What is Selank?
Selank is a synthetic heptapeptide analogue of tuftsin, designed for research into neuropsychotropic, anxiolytic, immunomodulatory, and cognitive modulation effects. It combines the amino acid sequence of tuftsin with an added Pro–Gly–Pro (PGP) tripeptide to improve metabolic stability and duration of action.

Chemical Structure of Selank
Selank features the amino acid sequence Thr–Lys–Pro–Arg–Pro–Gly–Pro (TKPRPGP), with a molecular formula of C₃₃H₅₇N₁₁O₉ and a molecular weight of approximately 751.9 Da, listed in PubChem (CID 11765600). It retains key tuftsin-receptor interaction motifs while improving metabolic resistance via the C-terminal PGP extension.

What Are the Effects of Selank?
- Anxiolytic and Nootropic Activity: Rodent studies and clinical trials show rapid anxiety reduction and cognitive improvement without sedative or dependency effects typical of benzodiazepines.
- GABAergic System Modulation: Selank induces gene expression changes in GABAergic neurotransmission in rat brain cortex, indicating allosteric modulation of GABAA receptors.
- Immunomodulation & Neuroprotection: Studies report decreased pro-inflammatory cytokine IL‑6, normalization of the Th1/Th2 balance, and enhanced expression of brain-derived neurotrophic factor (BDNF).
- Serotonin and Enkephalin Regulation: Selank alters monoamine levels (including serotonin) and inhibits enkephalin-degrading enzymes, supporting neuroprotective and mood-stabilizing pathways.
- Stroke & Alcohol Withdrawal: Preclinical models show anxiolytic effects during alcohol withdrawal and neuroprotective benefits in ischemic conditions.
